The idea to conduct bioinformatics and computational analysis research of Nitric oxide synthase genes came from my observations of patients who attended a sickle cell anemia clinic who came with repeated attacks of stroke, repeated infections, recurrent vaso-occlusive crisis and other complications. All these let me think and search about what is common in all these phenotypic presentations?. From reviewing, I found there are three human isoforms of NOS in immune cells, brain and epithelium that I thought may be a major risk if all of them were truncated. I did the analysis and SIFT and PolyPhen predicted six SNPs in nNOS, iNOS, and seven SNPs in eNOS genes as damaging and more researches are warented to figure the associations. My future vision in this topics is to move to epigenetic level of regulations of this genes especially in neurological diseases ad metabolic ones, aiming to find the key points and better understanding of our body as an integrative and interconnected systems.
